摘要
研究旨在揭示卵泡抑素(FST)基因在藏鸡不同组织及发育阶段中的表达特性。选取1、42、70、120、150日龄藏鸡公母各5只,分别采集垂体、下丘脑、胸肌、腿肌组织样本,利用qRT-PCR技术,对FST基因在所选组织中的表达水平进行测定,构建组织表达谱及时序表达谱并进行分析。结果显示:FST基因主要在藏鸡垂体组织中表达,发育前期公鸡中的表达水平较高,发育后期母鸡中的表达水平高于公鸡;与下丘脑、胸肌以及腿肌时序表达特性不同,FST基因在藏鸡垂体组织中的时序表达特性具有性别差异。研究结果为进一步了解FST基因的功能及物种多样性提供了参考,也为藏鸡的遗传育种提供了一定的理论基础。
        The aim of this study was to reveal the expression characteristics of follistatin(FST) gene in different tissues and developmental stages of Tibetan chicken. The pituitary, hypothalamus, breast muscle and leg muscle were obtained from Tibetan chickens of 1, 42, 70, 120 and 150 days of age(5 cocks and 5 hens at each stage), respectively.Using qRT-PCR technology detected the expression level of FST gene, and tissue expression profiles and time series expression profiles were constructed and analyzed. The results showed that FST gene was mainly expressed in the pituitary tissues of Tibetan chickens, the expression level in cocks was higher than that in hens in the pre-developmental,the opposite of late development; The temporal expression characteristics of FST gene in pituitary tissue had male-female differences, which was different from expression in other tissues. This study provided references for further understanding of the function and species diversity of FST gene, and also laid a theoretical foundation for the genetic breeding of Tibetan chicken.
