中药补肾解毒方对小鼠辐射后机体免疫功能的影响

摘要
目的观察中药补肾解毒方对小鼠辐射后机体免疫功能的影响。方法 90只无特定病原体级健康雄性小鼠随机分为空白对照组、辐射模型组、氨磷汀组、中药预防组、中药防治结合组,每组18只。除空白对照组外,其他4组小鼠接受一次性60Coγ-射线全身照射,吸收剂量为2 Gy。中药预防组和防治结合组照射前均采用补肾解毒方灌胃,中药防治结合组照射后继续采用补肾解毒方灌胃。均于照射后第1、7、14天取材,每组每次6只,眼球取血检测血清白细胞介素12(IL-12)、γ-干扰素(γ-IFN)含量。结果辐射后第1天,氨磷汀组、中药防治结合组IL-12水平明显低于辐射模型组、中药预防组[(65±16)、(66±34) ng/L比(94±42)、(96±21) ng/L](均P <0. 05)。辐射后第7天,5组间IL-12水平差异均无统计学意义(均P> 0. 05)。辐射后第14天,中药预防组IL-12水平明显低于空白对照组[(67±31) ng/L比(95±38) ng/L],中药防治结合组IL-12水平[(94±15) ng/L]明显高于中药预防组(均P <0. 05)。辐射后第1天,辐射模型组、氨磷汀组、中药预防组、中药防治结合组γ-IFN水平均明显低于空白对照组[(67±13)、(51±8)、(77±14)、(80±13) g/L比(106±32) g/L],中药预防组、中药防治结合组明显高于氨磷汀组(均P <0. 05)。辐射后第7天,空白对照组γ-IFN水平明显低于其他4组[(66±33) g/L比(101±13)、(98±14)、(108±9)、(122±26) g/L](均P <0. 05)。辐射后第14天,5组间γ-IFN水平比较差异均无统计学意义(均P> 0. 05)。结论补肾解毒方防治结合的给药方式较单纯辐射前预防给药效应明显持久,且可使机体IL-12、γ-IFN含量达到甚至略高于健康机体内含量水平,有效、持久、安全地增强了机体自身的免疫调节功能。
        Objective To analyze the effect of Chinese herbs tonifying-kidney and detoxification prescription on immunologic function in mice after radiation. Methods Ninety specific pathogen free healthy male mice were randomly divided into blank control group,model control group,amifostine group,pretreatment with traditional Chinese medicine group( TCM prevention group),combination of prevention and cure with traditional Chinese medicine group( TCM combination group),with 18 mice in each group. The mice were treated with 2 Gy60 Coγ-radiation except in blank control group. TCM prevention group and TCM combination group had gastric administration of tonifying-kidney and detoxification prescription before γ-radiation,and TCM combination group continued the intervention after γ-radiation. Eyeball blood was extracted in respectively 6 mice in each group on the 1,7,14 days after radiation to test levels of serum interleukin-12( IL-12) and γ-interferon( γ-IFN). Results On the 1 st day after radiation,IL-12 levels in amifostine group,TCM combination group were significantly lower than those in model control group and TCM prevention group[( 65 ± 16),( 66 ± 34) ng/L vs( 94 ± 42),( 96 ± 21) ng/L]( all P < 0. 05). On the 7 th day,there was no significant difference of IL-12 level among the 5 groups( P > 0. 05). On the 14 th day,IL-12 level in TCM prevention group was significantly lower than that in blank control group[( 67 ±31) ng/L vs( 95 ± 38) ng/L]and it in TCM combination group[( 94 ± 15) ng/L]was significantly higher than that in TCM prevention group( both P < 0. 05). On the 1 st day after radiation,γ-IFN levels in model control group,amifostine group,TCM prevention group and TCM combination group were significantly lower than those in blank control group[( 67 ± 13),( 51 ± 8),( 77 ± 14),( 80 ± 13) g/L vs( 106 ± 32) g/L]; the levels in TCM prevention group and TCM combination group were significantly higher than that in amifostine group( P < 0. 05). On the 7 th day,γ-IFN level in blank model control group was significantly lower than that in the other 4 groups[( 66 ± 33) g/L vs( 101 ± 13),( 98 ± 14),( 108 ± 9),( 122 ± 26) g/L]( P < 0. 05). On the 14 th day,there was no significant difference of γ-IFN level among 5 groups( P > 0. 05). Conclusion Combination of preventive and curative therapy with Chinese medicine tonifying-kidney and detoxification prescription shows an effective and safe regulatory function in protecting the immunity of mice treated with radiation; serum levels of IL-12 and γ-IFN can be persistently improved and recover to normal levels.
