摘要
引物的设计是决定PCR反应能否特异性及高效地扩增的要素之一。双启动寡聚核苷酸(dual priming oligonucleotide,DPO)引物,是一种包含两个独立的特异性引物区域和寡聚次黄嘌呤(poly I)碱基桥连结构的设计方法。近年来,基于双启动寡聚核苷酸引物的PCR技术被广泛应用于临床医学检验、食源性病原微生物检验及动植物病毒检验检疫等领域,检验结果符合检验检疫行业标准。与常规PCR引物相比较,双启动寡核苷酸引物具有特异性更强,实用性良好等优点。本综述系统地综述了双启动寡聚核苷酸引物的设计原理及DPO-PCR技术在检验检疫方面的研究进展并探讨其应用价值。
        Design of primers is one of the factors that determine whether PCR can be amplified efficiently and specifically. Dual priming oligonucleotide primer is a design method, which contains two separate specific primer domains and poly I bases joined by the bridge construction. The PCR technique with DPO primers is widely applied in the fields of clinical medical examination, food borne pathogenic microorganism inspection and inspection and quarantine of plant and animal viruses in recent years, and the test results meet the standards for inspection and quarantine. Compared with the conventional PCR primers, the specificity of the dual priming oligonucleotide primers are stronger and more practical. In this paper, the design principle of dual priming oligonucleotide primers and the research progress of DPO-PCR on inspection and quarantine were systematically reviewed, as well as its application value.
