摘要
鸡传染性法氏囊超强毒株(vvIBDV)可引起鸡群高死亡率及严重的免疫系统损伤,给养禽业带来巨大的经济损失。为探讨泰山松花粉多糖(TPPPS)体外抗vvIBDV感染鸡胚成纤维细胞(DF-1)作用机制,以DF-1细胞为宿主细胞,利巴韦林(Ribavirin)为抑病毒对照药物,从TPPPS对病毒的直接灭活作用、阻滞作用、抑制吸附及其复制4个方面探讨TPPPS抗vvIBDV机理。结果显示,在无毒质量浓度范围内TPPPS对vvIBDV的最高抑制率为92.4%,Ribavirin的最高抑制率仅为76.7%;在阻滞试验和抑病毒吸附试验中最高抑制率分别达到91.85%和78.36%;在抑制病毒复制试验和直接灭活病毒试验中抑制率很低,表明TPPPS不能抑制病毒复制和直接灭活病毒。结果表明,TPPPS在体外可有效抑制vvIBDV对DF-1细胞的感染,推测其抗病毒机制是TPPPS预处理DF-1细胞从而结合了病毒颗粒在细胞表面的吸附位点及病毒与多糖直接相互作用抑制病毒吸附细胞,或两种作用同时存在。提示TPPPS可作为预防及治疗超强鸡传染性法氏囊病病毒感染的药物。
        Very virulent infectious bursal disease virus(vvIBDV)can cause high mortality and severe immunosuppression of chickens and bring huge economic losses to poultry industry.To investigate the antivital mechanism of Taishan Pinus massoniana pollen polysaccharide(TPPPS)on very virulent infectious bursal disease virus infected DF-1 cells,DF-1 cells were used as host cells,ribavirin as an antiviral control drug,the antivital mechanism of TPPPS was discussed from the aspects of TPPPS direct inactivation of virus,blocking effect,inhibition of adsorption and its replication.The result indicated that the inhibition rate of TPPPS against vvIBDV is up to 92.4% within non cytotoxic concentration,ribavirin's highest inhibition rate is only 76.7%.The inhibition rate separately are up to 91.85%and 78.36%in blocking effect and inhibiting adsorption assay.The inhibition rate is very low in the replication inhibition assay and direct inactivation assay,indicating that TPPPS can not inhibit the virus replication and direct inactivation.The study showed that Taishan pine pollen polysaccharide had good anti-vvIBDV activity in vitro,deducing that TPPPS may combine with the viral particles of the adsorption site at the cell surface and TPPPS inhibit virus adsorption cells by interaction,or both effects simultaneously.Suggesting that TPPPS can be used as a drug for the prevention and treatment of infectious bursal disease.
