龙血竭胶囊对SD大鼠放射性皮肤损伤防护作用及机制的初步研究

摘要
目的:探讨龙血竭对SD大鼠皮肤放射性损伤的防护作用及可能机制。方法:100只雄性SD大鼠,随机数字表法分为5个组:空白对照组、龙血竭对照组、照射模型组、比亚芬治疗组和龙血竭治疗组,每组20只,比亚芬和龙血竭于电子线照射后外用于背部皮肤,观察照射后3天大鼠皮肤组织病理改变,血清和皮肤组织丙二醛(MDA)含量,超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、Na~+-K~+-ATP酶和Ca~(2+)-Mg~(2+)-ATP酶活性,Western Blot方法检测皮肤组织Bcl-2、Bax、Caspase-3蛋白表达水平。结果:龙血竭胶囊可以降低皮肤和血清MDA的含量,增加GSH-Px和SOD的活性,增加皮肤Bcl-2蛋白水平,降低Bax和活化的Caspase-3蛋白水平,对ATP酶活性无影响。结论:龙血竭胶囊对大鼠放射性皮肤损伤有防护作用,其机制可能为提高机体的抗氧化酶和抑制细胞凋亡。
        Objective:To explore the possible mechanism of the prevention of Longxuejie capsules in radiomermatitis using a SD rat model.Methods:One-hundred male SD rats were randomly divided into five groups:Blank control group,Longxuejie group,radiation group,biafine+radiation group and Longxuejie+radiation group.Longxuejie or Biafine was applied to the back ski after electron beam irradiation.At 3d after treatment,skin tissue and serum were collected to observe pathological changes,test activities SOD,GSH-Px,Na~+-K~+-ATPase and Ca~(2+)-Mg~(2+)-ATPase as well as the content of MDA,and detect protein expression of Bcl-2,Bax and caspase 3.Results:Longxuejie capsules could reduce the content of MDA in tissue,increased the activities of GSH-Px and SOD,enhanced the expression of Bcl-2 protein level,but reduced the levels of Bax and cleaved Caspase-3.It has no effect on ATPase.Conclusions:Longxuejie capsules could protect rat skin from radiation injury probably by promotion the antioxidant enzymes and inhibition apoptosis.
