针刺对营养性肥胖小鼠肠黏膜TLR1/TLR2基因的良性调控作用

摘要
目的:探究针刺对营养性肥胖小鼠肠黏膜TLR1/TLR2基因的良性调控作用。方法:100只C57BL/6小鼠,采用随机数字表随机选取10只为正常组,余下饲养高脂饮食8周,复制营养性肥胖鼠30只,随机均分成模型组、针刺14d组及针刺28d组,针刺组针刺中脘、关元、天枢、足三里,得气后留针10min,观察比较各组小鼠体质量、Lee’s指数、血清脂肪因子[瘦素(LP)、脂联素(ADPN)]、炎性因子[白介素-6(IL-6)、白介素-10(IL-10)及肿瘤坏死因子α(TNF-α)]含量变化及肠黏膜组织中TLR1/TLR2基因的表达差异。结果:模型鼠体质量、Lee’s指数、LP、ADPN、肠组织炎性因子(IL-6、IL-10、TNF-α)及TLR1及TLR2蛋白分布密度及基因表达均显著高于正常组(P<0.01),针刺后显著下降(P<0.05,P<0.01),针刺28d组疗效优于针刺14d组(P<0.05)。结论:针刺治疗肥胖,调整肠黏膜的免疫状态,下调关键炎性调控基因TLR1/TLR2的表达,达到消除慢性炎症治疗肥胖的目的。
        Objective: To investigate the effects on the regulation of TLR1/TLR2 gene in intestinal mucosa of obese mice by using acupuncture. Methods: Ten of 100 C57 BL/6 mice were randomly selected as normal group, and the others were fed with high-fat diet for 8 weeks to establish 30 obese mice. The obese mice were randomly divided into model group,acupuncture 14 days group and acupuncture 28 days group by random number table, and acupuncture group selected Zhongwan(CV12), Guanyuan(CV4), Tianshu(ST25), Zusanli(ST36), and then needled for 10 min. Observed the difference of body weight,Lee's index, and the content of LP, ADPN, IL-6, IL-10 and TNF-α in mice and the expression of TLR1/TLR2 in intestinal mucosa. Results: Many parameters of the body weight, Lee's index, LP, ADPN, IL-6, IL-10, TNF-α, and gene expression of TLR1 and TLR2 protein were higher than those of the normal group(P<0.01), however, they were decreased significantly after acupuncture(P<0.05, P<0.01). Moreover, the acupuncture 28 days group was superior to the acupuncture 14 days group(P<0.05). Conclusion: Acupuncture treating obesity can adjust the immune status of intestinal mucosa and down-regulate the expression of key inflammatory regulatory gene TLR1/TLR2, and it can eliminate chronic inflammation to reach the goal of obesity treatment.
