摘要
运用改良石蜡切片法制备了金针菇(Flammulina filiformis)子实体不同发育时期的切片,观察其菌褶和菌柄的细胞结构。结果表明,以柠檬烯为透明剂,金针菇子实体组织结构清晰、完整、无皱缩。观察不同发育时期的金针菇子实体组织切片发现,金针菇原基内部有呈菌丝无规则扭结状态的菌盖结构,随着子实体不断发育,菌盖下端有菌褶结构分化,随后菌柄结构随菌盖发育而进一步分化,菌柄组织细胞状纵向紧密排列,横切面有维管束状细胞呈外紧密内疏松状排列且其间有孔洞出现,菌褶菌髓部呈菌丝与球泡状细胞共同存在的异型分布结构。
        The improved paraffin method was used to prepare sections of Flammulina filiformis fruiting bodies at different developmental stages,and to observe cell structure of gills and stipes.The results showed that F.filiformis fruiting body section was clear,intact and wrinkle-free with limonene as clearing agent.Observation of tissue sections of F.filiformis fruiting bodies showed that there was a pileus structure with irregularly twisted mycelium in primordium.With the development of fruiting body,the gills differentiated in the bottom of pileus; after that,the stipe structure was further differentiated with the development of pileus.The stipe cells were longitudinally closely aligned in F.filiformis.Viewing from the cross section,vascular bundle cells were arranged as compacting inside and loosening outside with cavities in it.A heteromorphic structure was observed,where mycelia and bulliform cells coexisted in gill trama.
