摘要
以鞑靼忍冬的成熟种胚作为外植体,建立再生植株体系,结果表明:成熟种胚采用2%的NaClO溶液处理时,最佳消毒处理时间为6 min,存活率为75%。诱导愈伤组织以及诱导不定芽的最佳培养基为MS+2.0 mg/L 6-BA+0.1 mg/L NAA,愈伤组织诱导率为74.85%;不定芽诱导率为69.35%。增殖的最佳培养基为MS+2.0 mg/L 6-BA+0.3 mg/L NAA,增殖率为76.58%。诱导生根最佳培养基为1/2 MS+0.5 mg/L IBA,生根率为76.45%,平均根长为4.25 cm。通过组织培养再生植株,对其快速繁殖、种质资源保存和改良及规模化生产利用以及工厂化育苗技术研究提供理论基础。
        In this paper, the mature embryo of tatar honeysuckle was used as explants to establish plant regeneration system. The results showed that when the mature embryo was treated with 2% NaClO solution, the best disinfection time was 6 min and the survival rate was 75%. The best medium for callus and adventitious bud induction was MS+2.0 mg/L 6-BA+0.1 mg/L NAA, with the callus induction rate of 74.85%, and adventitious bud induction rate of 69.35%. The best medium for proliferation was MS +2.0 mg/L 6-BA+0.3 mg/L NAA, and the proliferation rate was 76.58%. The best medium for root induction was 1/2 MS+0.5 mg/L IBA, rooting rate was76.45%, and the average root length was 4.25 cm. Plants regenerated by tissue culture could provide theoretical basis for rapid propagation, conservation and improvement of germplasm resources, large-scale production and utilization, as well as industrial seedling technology research of tatar honeysuckle.
