中药复方糖克煎剂对2型糖尿病大鼠胰岛β细胞功能影响

摘要
目的:观察中药复方糖克煎剂对2型糖尿病(T2DM)大鼠胰岛β细胞功能的影响。方法:Wister大鼠随机分为正常组、模型组、糖克煎剂组、罗格列酮组,除正常组外其余各组均腹腔注射STZ联合高脂高糖饲料喂养建立2型糖尿病大鼠模型,正常组予等量无菌枸橼酸缓冲液注射。造模成功后糖克煎剂组予糖克煎剂10.8 g/(kg·d)灌胃,罗格列酮组予罗格列酮4.74 mg/(kg·d)进行灌胃,正常组和模型组均予蒸馏水5 mL/(kg·d)灌胃,连续4周,分别检测药物干预前后各组大鼠糖耐量、空腹胰岛素水平,并计算各组胰岛β细胞功能指数;利用铜试剂显色法检测各组药物干预后游离脂肪酸水平。结果:药物干预后糖克煎组和罗格列酮组HOMA-β均高于模型组(P<0.05),FFA水平均低于模型组(P<0.05)。结论:糖克煎剂显著降低T2DM模型大鼠血中FFA水平及使胰岛β细胞功能好转。
        Objective:To observe the effect of Chinese herbal compound Tangke Decoction on βcell function of pancreatic islet in type 2 diabetes mellitus(T2DM)rats. Methods:Wister rats were randomly divided into normal group,model group,Tangke Decoction group and Rosiglitazone group,the model of type 2 diabetic rats was established by intraperitoneal injection of STZ combined with high fat and high sugar feed feeding in all other groups except normal group,and the normal group was injected with the equivalent aseptic citrate buffer. After the successful model,the sugar-fried decoction group was 10.8g/(kg·d)Rosiglitazone group was irrigated with Rosiglitazone 4.74 mg/(kg·d),and the normal group and model group were infused with distilled water 5 mL/(kg·d)for 4 consecutive weeks,respectively,to detect the glucose tolerance of rats before and after drug intervention,fasting insulin level,and the function index of β cells of islet were calculated,and the level of free fatty acid was detected by copper reagent chromogenic method. Results:After drug intervention,both the Homa-β in the Tangke group and Rosiglitazone group were higher than the model group(P<0.05),and FFA levels were lower than the model group(P<0.05).Conclusion:The glucose decoction significantly reduced the FFA level in the blood of T2DM model rats and improved the function of islet β cells.
