摘要
建立了定性分析杜仲颗粒化学成分的超高效液相色谱-线性离子阱-静电场轨道阱组合式高分辨质谱(UPLC-LTQ-Orbitrap-MS)方法。杜仲颗粒采用甲醇超声提取,Thermo Scientific Syncronis C_(18)色谱柱(100 mm×2.1 mm,1.9μm)分离,以乙腈-0.1%甲酸溶液为流动相进行梯度洗脱。利用LTQ-Orbitrap-MS负离子模式采集,通过对照品比对及高分辨质谱数据解析,鉴定了25种化学成分,包括8种环烯醚萜类、8种黄酮类、5种苯丙素类、3种木脂素类化合物及1种非还原性双糖。该方法简便、快速、准确,可为进一步分析杜仲颗粒的药效物质基础提供依据。
        An ultra high-performance liquid chromatography coupled with linear ion trap-orbitrap mass spectrometric(UPLC-LTQ-Orbitrap-MS) method was developed for the qualitative analysis of main chemical constituents in Duzhong granules,a Chinese medicine.The sample was ultrasonically extracted with methanol,then separated on a Thermo Scientific Syncronis C_(18) chromatographic column(100 mm×2.1 mm,1.9 μm) by gradient elution with acetonitrile-0.1% formic acid as mobile phase.The data were collected by using LTQ-Orbitrap-MS in negative ion mode.According to the contrast of the reference standards and the accurate masses of molecules,a total of 25 compounds in Duzhong granules were identified,including 8 iridoids,8 flavonoids,5 phenylpropanoids,3 lignans and 1 non-reducing disaccharide.Results showed that,with the advantages of simplicity,rapidness and accuracy,the developed method was suitable for the identification of chemical constituents in Duzhong granules,which provided a reference for the pharmacological basis of further analysis of Duzhong granules.
