麻黄与淫羊藿拆分组分配伍对肾阳虚水肿模型的干预作用研究

摘要
目的:研究麻黄与淫羊藿拆分组分配伍对肾阳虚水肿模型大鼠的影响,探讨淫羊藿对肾阳虚水肿病症大鼠干预作用的物质基础。方法:用氢化可的松联合盐酸多柔比星复制肾阳虚水肿大鼠模型,按照3.75mg/kg体重腹腔注射氢化可的松,连续注射14d。于第1天以4mg/kg和第7天3.5mg/kg尾静脉注射盐酸多柔比星溶液。造模结束后,将造模成功大鼠分为模型组、麻黄247mg/kg组、淫羊藿218mg/kg组、麻黄+淫羊藿(247+218)mg/kg组、麻黄+淫羊藿多糖(247+16)mg/kg组、麻黄+淫羊藿非黄酮(247+106)mg/kg组、麻黄+淫羊藿大极性黄酮(247+14)mg/kg组、麻黄+淫羊藿小极性黄酮(247+10)mg/kg组,同时设立空白对照组,连续灌胃14d。给药结束后,测定尿蛋白、血浆白蛋白(ALB)、血清总蛋白(TP)、环磷酸腺苷(cAMP)、环磷酸鸟苷(cGMP)、三碘甲状腺原氨酸(T3)、甲状腺素(T4)、血清肌酐(Crea)、血清尿素氮(BUN)、三酰甘油(TG)、总胆固醇(CHOL)、雌二醇(E2)和睾酮(T)含量。结果:与模型组比较,麻黄使大鼠体重及肛温显著上升,Crea、BUN、尿蛋白含量显著性下降;淫羊藿使T水平显著性上升,cGMP、TG、尿蛋白含量下降、Crea、BUN水平显著性下降;麻黄+淫羊藿能增加大鼠体重,显著增加T水平,TG、CHOL、Crea、BUN含量下降,尿蛋白含量显著降低;麻黄+淫羊藿大极性黄酮组可以使大鼠体重、cAMP/cGMP显著上升,E2、T、TG、ALB和TP含量显著上升,使尿蛋白、cGMP、Grea、BUN含量显著下降;麻黄+小极性黄酮组可以使大鼠体重、cAMP、cAMP/cGMP和T3升高,E2、T升高,使尿蛋白、CHOL含量下降、Crea、BUN含量显著下降;麻黄+非黄酮组能升高大鼠肛温和E2含量,降低Crea、BUN含量;麻黄+多糖组能增加体重、肛温、T4和E2含量,并且使BUN含量降低。结论:麻黄+淫羊藿拆分组分配伍均能温补肾阳又能利水消肿,对肾阳虚水肿模型大鼠有良好的治疗效果,治疗效果为:麻黄+大极性黄酮组分>麻黄+小极性黄酮>麻黄+非黄酮>麻黄+多糖。淫羊藿中的大极性黄酮组分和小极性黄酮组分是利水消肿功效的主要物质基础。
        Objective: To study the effect of the single and combination of Herba Ephedra and Herba Epimedium on Shenyangxu Edema rats model,and to explore the material basis of Epimedium on Shenyangxu Edema model rats. Methods: Rat model of Shenyangxu Edema was established by hydrocortisone combined with doxorubicin hydrochloride. The hydrocortisone was intraperitoneally injected at the dose of 3. 75 mg/kg for14 days. The doxorubicin hydrochloride solution was intravenously injected on the 1 st and 7 th day,at the dose of 4 mg/kg and 3. 50 mg/kg respectively. After the model was established,all rats were divided into the model group,Ephedra group( 247 mg/kg),Epimedium group( 218 mg/kg),Ephedra + Epimedium group( 247 + 218 mg/kg),Ephedra + Epimedium polysaccharide group( 247 + 16 mg/kg),Ephedra+ Epimedium non-flavonoid group( 247 + 106 mg/kg),Ephedra + Epimedium macroporous flavonoid group( 247 + 14 mg/kg),Ephedra+ Epimedium low polarity flavonoid group( 247 + 10 mg/kg) and the control group. Rats in the drug-administered groups were given the corresponding drugs,and the rats in the control group and the model group were given an equal volume of distilled water for 14 days. After the administration,urine protein,plasma albumin( ALB),serum total protein( TP),cyclic adenosine monophosphate( cA MP),cyclic guanosine monophosphate( cG MP),triiodothyronine( T3),and thyroid gland were measured.( T4),also the contents of serum creatinine( CREA),serum urea nitrogen( BUN),triglyceride( TG),total cholesterol( CHOL),estradiol( E2) and testosterone( T) were determined. Results: Compared with the model group,body weight and anal temperature were increased,contents of CREA,BUN,and urine protein were significantly decreased in Ephedra group. In Epimedium group,T level was significantly increased,the levels of cG MP,TG,CREA,BUN and urine protein were decreased. In Ephedra + Epimedium group,the body weight index was increased and T level was significantly corrected. The contents of TG,CHOL,CREA and BUN were decreased and the content of urinary protein was significantly decreased. In the macroporous flavonoids of Ephedra and Epimedium group,body weight and cA MP/cG MP were significantly increased,the levels of E2,T,TG,ALB and TP were increased,the contents of BUN,urine protein and cG MP were decreased. In Ephedra + Epimedium low polarity flavonoids group,the body weight,cA MP,cA MP/cG MP and T3 were increased,levels of E2 and T were significantly increased,the content of urine protein CHOL was decreased,the content of CREA. BUN was significantly decreased. In Ephedra + non-flavonoids Epimedium group,rat anal temperature and E2 content were increased,contents of CRE A and BUN were decreased. In Ephedra + Epimedium polysaccharide group,the body weight,the anal temperature,T4 and E2 contents were increased,BUN content reduced. Conclusion:Single and combination of Ephedra and Epimedium prescription have the good therapeutic effect on rats with Shenyangxu and edema. The therapeutic effects are shown in order: Ephedra + macroporous Epimedium flavonoids > Ephedra + low polarity Epimedium flavonoids >Ephedra + non-flavonoids Epimedium > ephedra + Epimedium polysaccharide. The macroporous flavonoid component and the low polarity flavonoid component in Epimedium are the main material basis in Lishuixiaozhong effect.
