湖南省稻水象甲的遗传多样性及入侵扩散特点

摘要
为全面了解湖南省稻水象甲的入侵扩散特点,运用简单重复序列间区(ISSR)分子标记方法及对比COⅠ基因、ITS2片段的序列信息,探讨全省17个地理种群的遗传多样性和扩散模式。结果表明:湖南省稻水象甲种群COⅠ基因、ITS2片段的序列非常保守,不能提供有效的多态性位点信息,而ISSR的多态性信息非常丰富,其多态性位点百分率可达93.24%;基于ISSR的数据分析发现,湖南省稻水象甲的Nei’s基因多样性指数为0.363 0,各地理种群间基因分化系数为0.414 3,种群之间的遗传距离与地理距离无相关性。湖南省稻水象甲地理种群分化不符合地理隔离模式,以人为因素导致的扩散为主,自然扩散为辅。
        To understand the geographical range expansion of Lissorhoptrus oryzophilus(L. oryzophilus), a rice weevil,in Hunan province, inter–simple sequence repeat(ISSR) molecular markers, mitochondrial COⅠgene and ITS2 sequence were examined to investigate the genetic diversity and expansion model of 17 geographic populations. The results showed that mitochondrial COⅠ gene and ITS2 sequence were both conservative in different individuals, which was inapropriate for effective polymophism analysis. However, the percentage of polymorphic bands of ISSR of populations reaches to 93.24%. After further analyzing, we found that Nei's gene diversity index(H) was 0.363 0, and the coefficient of genetic differentiation(Gst) among populations was 0.414 3. There was no significant correlation between genetic distance and geographical distance. The morphological differentiation of L. oryzophilus is not in conformity with the geographical isolation mode in Hunan province, indicating the rapid spread of rice water weevil in Hunan province was mainly related to artificial transport and less related to natural factors.
