新塔花水提物对ApoE~(-/-)动脉粥样硬化模型小鼠血清中IL-6、IL-10表达的影响

摘要
目的:观察新塔花水提物对载脂蛋白E基因敲除(ApoE~(-/-))小鼠血清中促炎因子白细胞介素-6(IL-6)和抗炎因子白细胞介素-10(IL-10)表达的影响,探讨新塔花防治动脉粥样硬化(AS)的可能抗炎机制。方法:40只8周龄ApoE~(-/-)小鼠分为模型组,阿托伐他汀组,新塔花高、低剂量组,每组10只;同品系C57BL/6J小鼠10只为空白组。所有小鼠给予高脂饲料室温饲养。适应性喂养2周后,实验组给予一定剂量药物灌胃干预,空白组和模型组给予等量蒸馏水灌胃。灌胃12周后处死各组小鼠,采集标本,HE染色光镜下观察主动脉病理学变化,测定斑块面积(PA)与管腔面积(LA)之比;ELISA法检测血清中IL-6和IL-10含量。结果:模型组主动脉PA/LA比值显著大于空白组(P<0.01),新塔花高剂量组、阿托伐他汀组PA/LA比值则显著小于模型组(P<0.01)。模型组血清中IL-6、IL-10表达水平均高于空白组(P<0.01),新塔花高剂量组、阿托伐他汀组血清中IL-6表达水平均显著低于模型组(P<0.01),IL-10表达水平显著高于模型组(P<0.01)。结论:新塔花具有预防AS的作用,其机制可能与降低炎症因子IL-6和提高抗炎因子IL-10的表达水平,抑制炎性反应有关,并具有一定的药物剂量依赖性。
        Objective: To observe the effects of water extract of Ziziphora clinopodioides Lam. on the expression of interleukin-6(IL-6) and interleukin-10(IL-10) in ApoE~(-/-) atherosclerosis model mice, and to explore the possible mechanism of Ziziphora clinopodioides Lam. in the prevention and treatment of atherosclerosis(AS). Methods: Forty ApoE~(-/-) mice of 8 weeks' old were divided into model group, atorvastatin group, Ziziphora clinopodioides Lam. high-dose and low-dose group, with 10 mice in each group. Ten strains of C57 BL/6 J mice were in blank group. All mice were fed high-fat diet at room temperature. After 2 weeks of adaptive feeding, the experimental group was given a certain dose of drug intragastrically, and the blank and model group were given equal volume of distilled water. After 12 weeks of gavage, each group of mice was sacrificed and specimens were collected. HE staining was used to observe the pathological changes of the aorta, and the ratio of plaque area(PA) to lumen area(LA)was measured. The levels of IL-6 and IL-10 in serum were detected by ELISA. Results: The PA/LA ratio in the aorta of the model group was greater than that in the blank group(P<0.01). The ratio of PA/LA in Ziziphora clinopodioides Lam. high-dose group and atorvastatin group were smaller than that in the model group(P<0.01). The serum level of IL-6 and IL-10 in the model group was higher than that in the blank control group(P<0.01). The IL-6 expression level in the serum of the Ziziphora clinopodioides Lam. high-dose group and atorvastatin group was lower than that in the model group(P<0.01), while IL-10 expression levels were higher than the model group(P<0.01). Conclusion: Ziziphora clinopodioides Lam. has the effect of preventing atherosclerosis,and its mechanism may be related to the reduction of IL-6 and the increase of anti-inflammatory cytokine IL-10 expression level,inhibition of inflammatory response, and has a certain dose-dependence.
