摘要
目的建立虫草花中麦角甾醇的提取及含量测定方法。方法虫草花经甲醇超声提取后,用RP-HPLC测定。采用Agilent Eclipse C_(18)(2.1×50mm,1.8μm)色谱柱,流动相为甲醇-水(98∶2);流速为0.2mL·min~(-1);检测波长为280nm。结果麦角甾醇在5.56~556μg·mL~(-1)与峰面积呈良好的线性关系(r=0.9997);麦角甾醇的平均回收率为99.70%(RSD=0.48%,n=9)。结论该方法快速、简便,结果准确、可靠。
OBJECTIVE To extract ergosterol from Cordyceps militaris L. Link and determine its content. METHODS The optimal extracting condition was optimized by orthogonal design method,and the content of ergosterol was determined by RP-HPLC. RESULTS The ergosterol presented a good linear correlation in the range of 5. 56-556μg·mL-1( r = 0. 9997). The average recovery was 99. 70%( RSD = 0. 48%,n = 9). CONCLUSION This extracting method is rapid and precise,with accurate and reliable results.
