不同产地白蔹中总酚酸以及没食子酸、原儿茶醛、儿茶素和白藜芦醇的含量测定

摘要
目的建立白蔹药材中总酚酸的含量测定方法,比较不同产地白蔹中总酚酸以及没食子酸、原儿茶醛、儿茶素和白藜芦醇的含量。方法超声波提取法提取白蔹总酚酸,采用紫外-可见分光光度法,以没食子酸为对照品,0.6%FeCl_3:0.9%K_3[Fe(CN)_6](1∶1)混合溶液为显色剂,在735 nm波长处进行测定;采用HPLC法测定不同产地没食子酸等4个成分的含量。结果总酚酸在0.1696~1.3568 g/m L(r=0.9993)浓度范围内线性关系良好,平均回收率为104.0%,RSD为2.713%(n=6)。不同产地白蔹中总酚酸含量在2.51%~3.73%范围内,以浙江产白蔹含量最高;不同产地白蔹的4个成分中儿茶素含量最高,白藜芦醇和原儿茶醛的含量较低,各个产地间含量差异较大。结论建立的总酚酸含量测定方法简单快捷、稳定、重现性好,该研究为完善现有白蔹质量评价体系及后续产品的开发提供参考。
        Objective To establish a method for the determination of total phenolic acid in,and compare the contents of total phenolic acid and gallic acid,protocatechin aldehyde,catechin and resveratrol in different producing areas. Methods 0.6%Fe Cl3∶0.9%K3[Fe(CN)6](1∶1)mixed solution was used as a colorant to determine total phenolic acid by ultrasonic extraction. The content of gallic acid in different habitats was determined by HPLC method by using the mixed solution of gallic acid and 0.6%Fe Cl3∶0.9%K3[Fe(CN)6](1∶1)as color reagent. Results The total phenolic acids had good linear relationship in the concentration range of 0.1696~1.3568 ug/m L(r=0. 9993),with an average recovery of 104% and RSD of 2.713%(n=6). The content of total phenolic acids in Ampelopsis japonica from different habitats was in the range of2.51~3.73%,with the highest content in Zhejiang,and the content of catechin was the highest,the content of resveratrol and protocatechualdehyde was lower. Conclusion The determination method of total phenolic acid content is simple,fast,stable and reproducible,which provides a reference for the improvement of the existing quality evaluation system and the development of subsequent products.
