摘要
目的运用静息态功能磁共振成像(rs-fMRI)技术,探讨睡眠剥夺(SD)对健康成年男性情绪功能网络的影响及其可能的神经机制。方法采用自身前后对照设计,30名健康成年男性分别于正常睡眠后及36 h SD后进行rs-fMRI扫描。选取双侧杏仁核为种子区域,采用功能连接的分析方法,比较SD前后双侧杏仁核种子点与全脑功能连接的变化情况。结果 SD导致内侧前额叶、楔前叶与杏仁核功能连接增强(P<0.05);而与丘脑、岛叶之间的功能连接减弱。与36 h SD前相比,SD后POMS得分明显升高,两者差异有统计学意义(t=9.436;P=0.007);双侧杏仁核与内侧前额叶皮质、丘脑间的功能连接与心境状态量表(POMS)得分呈正相关(r=0.614,0.492;P=0.013,0.027;r=0.537,0.453;P=0.021,0.033)。结论 36 h SD后大脑负责情绪调控的双侧杏仁核区功能连接显著改变,主要表现为与内侧前额叶皮质功能连接增强,而与双侧丘脑功能连接减弱;这可能是SD后负性情绪增加,正性情绪下降的重要神经机制。
        Objective Functional connectivity analysis was used to analyze the effects of sleep deprivation on the emotional functioning network in healthy adult men under resting state fMRI. Methods Amygdala were selected as seed points. Using resting state fMRI and functional connectivity analysis to compare the whole brain functional connectivity patterns of 30 young healthy male volunteers in the rested wakefulness state and sleep deprivation state. Results Sleep deprivation resulted in a significant increase between the medial prefrontal cortex,anterior wedge and amygdala(P<0.05). The functional connectivity was significantly decreased between amygdala,thalamus and insular.Compared with rested wakefulness state the total score of profile of mood states(POMS) was significantly higher in the sleep deprivation state.The connection between amygdala and the medial prefrontal cortex,thalamus showed positive correlation with the total score of POMS(r=0.614,0.492; P=0.013,0.027; r=0.537,P=0.021; r=0.453,P=0.033). Conclusion SD has a significant effect on amygdala functional connectivity.The functional connectivity between amygdala and the medial prefrontal cortex is significantly increased while that between amygdala and thalamus is decreased after 36 hours SD.This findings may be the essential neural mechanism of negative emotion raised,positive emotion decline induced by sleep deprivation.
