摘要
目的探讨白花延龄草水提液对肝纤维化作用及其作用机制。方法腹腔注射四氯化碳(CCl_4)构建雄性C57鼠肝纤维化模型,给药A组(白花延龄草水提液0. 5 g/kg)、给药B组(白花延龄草水提液1 g/kg)、阳性对照组(扶正化瘀胶囊0. 75 g/kg)分别进行灌胃给药,采用生化方法检测血清中的丙氨酸氨基转移酶(ALT)和天门冬酸氨基转移酶(AST)及肝脏组织中的超氧化物歧化酶(SOD)、脂质氧化终产物丙二醛(MDA)含量;苏木精-伊红(HE)染色法检测肝纤维化的变化;蛋白质印迹(Western Blot)法检测转化生长因子-β(TGF-β)蛋白表达水平。结果给药组和阳性对照组均能显著抑制CCl_4诱导的小鼠肝纤维化程度,与模型组相比,给药组和阳性对照组血清ALT,AST水平及肝脏组织中MDA水平明显降低(P <0. 01),SOD显著提高(P <0. 01);与模型组相比,给药组和阳性对照组TGF-β蛋白表达具有显著的抑制作用(P <0. 01)。结论白花延龄草水提液对肝纤维化有显著的改善作用。
        Objective To investigate the protective effect of water extract of Trillium kamtschaticum on liver fibrosis. Methods The liver fibrosis model of male C57 mice was established by intraperitoneal injection of CCl_4. Group A(0. 5 g/kg water extract of Trillium kamtschaticum) and group B( 1 g/kg water extract of Trillium kamtschaticum) and positive control group( 0. 75 g/kg Fuzheng Huayu Capsules) were given intragastric administration respectively. The levels of alanine aminotransferase( ALT) and aspartate aminotransferase( AST) in serum and the content of superoxide dismutase( SOD) and malondialdehyde( MDA) in liver tissue were detected by biochemical method. The change of liver fibrosis was measured by HE stain method. The expression level of transforming growth factor-β( TGF-β)protien was detected by Western Blot method. Results Both the drug group and the positive control group could significantly inhibit CCl_4-induced liver fibrosis in mice. Compared with the fibrosis model group,the levels of ALT,AST in serum and MDA in liver tissue in the drug group and the positive control group were significantly lower( P < 0. 01),while the levels of SOD were significantly higher( P < 0. 01). Compared with the model group,the expression level of TGF-β protein was significantly inhibited in the drug group and the positive control group( P < 0. 01). Conclusion The water extract of Trillium kamtschaticum has significant effect on improving liver fibrosis.
