摘要
目的探讨复方丹参滴丸对短暂性脑缺血大鼠脑组织中Nod样受体蛋白3(NLRP3)、胱天蛋白酶1(Caspase-1)、白细胞介素(IL)-1β和IL-18表达的影响。方法经改良Longa线栓法制作大鼠短暂性大脑中动脉闭塞(tMCAO)模型。将SD雄性大鼠随机分为假手术组、模型组和实验组。于大鼠苏醒后1 h,实验组予以145 mg·kg~(-1)·d~(-1)复方丹参滴丸,qd,灌胃。模型组和假手术组均予以等体积0. 9%NaCl。用苏木素-伊红(HE)染色法和尼氏染色法观察脑组织病理形态的改变,用过氧化物酶(MPO)免疫组化染色法检测炎性细胞浸润,用实时荧光定量聚合酶链式反应法检测NLRP3、Caspase-1、IL-1β和IL-18mRNA的表达水平,用酶联免疫吸附法检测脑组织中细胞因子IL-1β和IL-18的蛋白表达。结果假手术组、模型组和实验组的神经元数量分别为(614. 4±74. 2),(191. 6±35. 4)和(352. 3±29. 5) cell·mm-2,MPO免疫组化评分分别为0. 08±0. 29,7. 83±2. 21和4. 00±1. 13,实验组上述指标与模型组比较,差异均有统计学意义(均P <0. 01)。与模型组比较,实验组1,3 d时的NLRP3、IL-1β、IL-18 mRNA表达水平均显著下调,差异均有统计学意义(P <0. 01或0. 05),1 d时Caspase-1 mRNA表达水平显著下调,差异有统计学意义(P <0. 01)。与模型组比较,实验组1,3 d时IL-1β蛋白表达水平均显著下调,1 d时IL-18蛋白表达水平显著下调,差异均有统计学意义(P <0. 01或P <0. 05)。结论复方丹参滴丸能改善短暂性脑缺血模型大鼠脑组织损伤,该作用与抑制NLRP3介导的炎症反应有关。
        Objective To research the effects of compound Danshen dripping pills on the expressions of Nod-like receptor 3( NLRP3),cysteinyl aspartate specific proteinase 1( Caspase-1),interleukin( IL)-1β and IL-18 in the brain of transient ischemic rats. Methods Transient middle cerebral artery occlusion( t MCAO) model was established by modified Longa suture method. SD male rats were randomly divided into sham-operation,model and experimental groups. After one hour of awakening,the experimental group was given 145 mg·kg~(-1)·d~(-1) compound Danshen dropping pills by gavage,qd. Both the model group and the sham-operation group were given an equal volume of 0. 9% NaCl.Hematoxylin-eosin( HE) staining and Nissl staining were used toobserve the pathological changes of brain tissue. Inflammatory cell infiltration was detected by peroxidase( MPO) immunohistochemical staining. The expression levels of NLRP3,Caspase-1,IL-1β and IL-18 mRNA were detected by real-time fluorescence quantitative polymerase chain reaction. The protein expressions of cytokines IL-1β and IL-18 in brain tissue were detected by enzyme-linked immunosorbent assay. Results The number of neurons in sham-operation group,model group and experimental group were( 614. 4 ± 74. 2),( 191. 6 ± 35. 4) and( 352. 3 ± 29. 5) cell·mm-2,respectively. The MPO immunohistochemical scores were 0. 08 ± 0. 29,7. 83 ± 2. 21 and 4. 00 ± 1. 13,the above indexes of experimental group had statistically significant differences compared with those of model group( all P < 0. 01). Compared with model group,the mRNA expression levels of NLRP3,IL-1β and IL-18 were significantly down-regulated at 1 d and 3 d in experimental group( P < 0. 01 or P < 0. 05),and the expression level of Caspase-1 mRNA was significantly down-regulated at 1 d( P < 0. 01). Compared with model group,IL-1β protein expression level was significantly down-regulated at 1 d and 3 d,and IL-18 protein expression was significantly down-regulated at 1 d( P < 0. 01 or P < 0. 05). Conclusion Compound Danshen dripping pills can improve the brain tissue damage of transient ischemia model rats,this effect is related to the inhibition of NLRP3 mediated inflammatory reaction.
