摘要
目的:分析冠心病患者外周血内皮祖细胞(EPCs)中sirtuins蛋白的表达。方法:选取经冠状动脉造影确诊为冠心病的患者37例(研究组)及正常对照35例(对照组),分别采集外周血分离EPCs细胞。利用qRTPCR检测EPCs细胞中sirt1~sirt7mRNA的表达,利用Western blot检测sirt1~sirt7蛋白的表达,利用酶联免疫吸附试验检测血清血管内皮生长因子(VEGF)、高敏C反应蛋白(hs-CRP)、同型半胱氨酸(Hcy)、血管紧张素Ⅱ(AngⅡ)的表达。结果:与对照组比较,研究组的年龄等指标无明显差异。研究组EPCs数量和VEGF水平明显低于对照组,而hs-CRP、Hcy、AngⅡ水平则明显高于对照组。研究组EPCs中sirt1、sirt3、sirt4、sirt6mRNA和蛋白表达水平显著低于对照组,而sirt2、sirt5、sirt7的表达水平则变化不明显。Sirt1和sirt6mRNA水平与VEGF水平具有正相关关系(sirt1:r2=0.7383,P<0.001,sirt6:r2=0.5576,P<0.001),与hs-CRP水平具有负相关关系(sirt1:r2=0.6568,P<0.001,sirt6:r2=0.5364,P<0.001),与Hcy及AngⅡ水平则无明显相关关系。结论:sirtuins家族蛋白在冠心病患者外周血EPCs中差异性表达,其中sirt1和sirt6表达水平与冠心病相关指标显著相关。
        Objective:To explore the expression of sirtuins in endothelial progenitor cells(EPCs)from peripheral blood of patients with coronary artery disease(CAD).Method:Patients diagnosed with CAD by coronary angiography(n=37,research group)and normal controls(n=35,control group)were enrolled in this study.Peripheral blood were collected and EPCs were then isolated.The expression of sirt1~sirt7 mRNA was analyzed using qRT-PCR method.The expression of sirt1~sirt7 protein was evaluated using Western blot assay.Moreover,the levels of serum vascular endothelial growth factor(VEGF),high-sensitivity C-reactive protein(hs-CRP),homocysteine(Hcy),and angiotensinⅡ(AngⅡ)were measured using enzyme linked immunosorbent assay(ELISA).Result:Compared with the control group,the indices like age in research group varied indifferently.EPCs numbers and VEGF levels in research group were significantly lower than control group,in contrast to hs-CRP,Hcy and AngⅡlevels,which were higher than control group.The expression of sirt1,sirt3,sirt4 and sirt6 mRNA and protein in EPCs from research group was significantly reduced compared with control group,while the expression of sirt2,sirt5 and sirt7 altered insignificantly.The expression of sirt1 and sirt6 mRNA was positively correlated with VEGF levels(sirt1:r2=0.7383,P<0.001,sirt6:r2=0.5576,P<0.001),negatively correlated with hs-CRP levels(sirt1:r2=0.6568,P<0.001,sirt6:r2=0.5364,P<0.001)and insignificantly correlated with Hcy and AngⅡlevels.Conclusion:Sirtuins are differentially expressed in EPCs from peripheral blood of CAD patients,with significant correlation between sirt1 and sirt6 expression and CAD related indexes.
