利用秋水仙碱诱导美洲黑杨2n花粉的初步研究

摘要
为了获得美洲黑杨三倍体植株,本试验采用二因素随机区组试验设计,对利用秋水仙碱诱导美洲黑杨2n花粉技术进行研究,结果表明:(1)水培3 d、4 d,5 d时,利用0.3%、0.5%、0.8%的秋水仙碱对美洲黑杨雄花芽进行处理均能得到一定比例的2n花粉;(2)在水培3 d时,利用0.5%的秋水仙碱处理美洲黑杨雄花芽,得到的2n花粉最多,为23.4%。
        In order to get the triploids of P. deltoid,the technique was researched to induce 2 n pollen of P. deltoid by colchicines by two-factor randomized block design. The results indicated that( 1) a certain proportion of 2 n pollen grains could be gotten from the male buds induced by cochicine solution with the concentration of 0. 3%,0. 5% and 0. 8%,under the condition of water culture for 3,4 and 5 days.(2)the proportion of 2 n pollen grains could be the highest when the male buds were treated by the cochicine solution of 0. 5% under the condition of water culture for 3 days,the percentage of A4( d ≧ 50) could be up to 1. 4%,the percentage of 2 n pollen( d ≧ 37. 5) could be up to 23. 4%.
