A molecular mode of action of Connexin43 mimetic peptide (Peptide5) is proposed. Peptide5 blocks Connexin43 hemichannel-mediated ATP release. Peptide5 acts on the extracellular loop two of Connexin43. Peptide5 sequence specificity is required for hemichannel block. The SRPTEKT motif is central to Peptide5 mechanism.
