A capillary zone electrophoresis(CZE)method has been established for a nalysis of the monosaccharide compositions of polysaccharides from Chinese herbRhizoma Phragmitis and Pollen Typhae.These monosaccharides derived with α-naphthylamine were detected by a diode array detector at 214nm.The five mono-saccharides rhamnose(Rha),xylose(Xyl),galactose(Gal),glucose(Glu),andarabinose(Ara)were simultaneously separated under the optimized CE conditionsof 50 mmol/L of sodium tetraborate buffer at pH 10.5 and 25 kV running voltage at25℃.The calibration plots were linear over a range from 5 to 200mg/L.The detec-tion limit(S/N=3)was 2.5mg/L for each monosaccharide.The relative standarddeviations of migration times and peak areas were<1.46% and 6.21%,respective-ly.The result showed that Rhizoma Phragmitis polysaccharides consist of Xyl,Glu,Ara,and Gal with molar ratio of 1.00:59.03:2.09:1.96;Pollen Typhaepolysaccharides consist of Rha,Xyl,Glu,Ara,and Gal with molar ratio of 1.71:1.00:9.30:6.88:1.24.
